In nature, it is found primarily in the outer layers of grains, yeast, peanuts, meat, animal organ offal, milk, and green leafy vegetables. It can be synthesized from tryptophan in the body, but with very low efficiency. Niacin can be synthesized by Escherichia coli and other bacteria in the intestines and then converted to niacinamide.

The main methods of synthesizing nicotinamide include chemical and biological methods. Chemical methods mainly include the synthesis of nicotinic acid from 3-methylpyridine and the synthesis of nicotinamide from nicotinic acid. The biological method is mainly through the reaction of 3-methylpyridine catalyzed by microorganisms to produce 3-cyanopyridine, and then 3-cyanopyridine is hydrolyzed to obtain nicotinamide.
